Solution

The Kd value, or partition coefficient, is a measure of how a protein distributes itself between the mobile phase (the liquid flowing through the column) and the stationary phase (the gel inside the column). It is calculated using the formula:

Kd = (Ve - V0) / (Vt - V0)

where: Ve = elution volume (the volume at which the protein comes out of the column) V0 = void volume (the volume at which molecules too large to enter the pores come out of the column) Vt = total pore volume (the volume of the gel inside the column)

In this case, the protein was eluted at 15 ml (Ve), the void volume is 5 ml (V0), and the pore volume is 1 ml (Vt).

However, the pore volume is not directly used in the formula. Instead, the total volume (Vt) is used, which is the sum of the void volume (V0) and the pore volume. In this case, Vt = V0 + pore volume = 5 ml + 1 ml = 6 ml.

Substituting these values into the formula gives:

Kd = (15 ml - 5 ml) / (6 ml - 5 ml) = 10 ml / 1 ml = 10

So, the Kd value of the protein is 10.
