In reverse phase chromatography, the stationary phase is made of a hydrophobic (water-repelling) material. This can be a silica gel that has been modified with R groups, which are long hydrocarbon chains. These chains interact with the sample components that are also hydrophobic, causing them to be retained on the stationary phase. The more hydrophobic the sample component, the longer it will be retained. This is the opposite of normal phase chromatography, where the stationary phase is polar and the mobile phase is non-polar.
