1. Selection of Bacteria: The Ames test uses a strain of the bacterium Salmonella typhimurium that carries mutations in the genes involved in histidine synthesis. This means the bacteria cannot produce their own histidine, an essential amino acid, and must rely on it being provided in their environment.

2. Exposure to the Test Substance: The bacteria are then exposed to the substance being tested. If the substance is a mutagen, it will cause mutations in the bacteria's DNA.

3. Incubation: The bacteria are then placed in an environment without histidine. Only those bacteria that have undergone a reverse mutation (which allows them to produce histidine again) will survive.

4. Observation: The number of colonies that grow is counted. A substance that causes more colonies to grow is considered more mutagenic.

5. Rat Liver Extract: The test is often run in the presence of a rat liver extract, which contains enzymes that can convert non-mutagenic substances into mutagens. This simulates the "activation" of certain substances in the body.
