This is an example of an indirect ELISA. In this type of ELISA, the test sample is added to a well coated with antigen. If the sample contains antibodies that recognize the antigen, they will bind to it. Then, a secondary antibody that recognizes human antibodies and is conjugated to an enzyme (in this case, HRP or horseradish peroxidase) is added. If the secondary antibody binds, the enzyme will convert a substrate (in this case, TMB or 3,3',5,5'-Tetramethylbenzidine) into a detectable signal. The signal is then stopped and read using a spectrophotometer.
