Adenosine deaminase (ADA) deficiency is a rare genetic disorder that is characterized by a lack of the enzyme adenosine deaminase. This enzyme is crucial for the breakdown of adenosine from food and for the turnover of nucleic acids in tissues. The lack of ADA leads to the accumulation of dATP, which inhibits ribonucleotide reductase and prevents DNA synthesis, leading to lymphocyte development failure and severe combined immunodeficiency (SCID).

Gene therapy for ADA deficiency involves the following steps:

  1. Isolation of lymphocytes from the patient's blood.

  2. In the laboratory, a functional ADA gene is inserted into these cells. This is usually done using a vector (often a virus that has been genetically altered to carry human DNA).

  3. The cells are then returned to the patient's body, where they start to produce the ADA enzyme.

This therapy has been successful in several patients, leading to a functional immune system. However, it is still considered experimental and is currently only used for patients who do not have a suitable bone marrow donor.
